The rail repair that needed less crew than anyone thought
First light sat cold on the rail yard. One short stretch of steel had been marked in paint, and a new piece waited beside it, cut to fit at both ends. The whole shift hung on opening that exact bit of track and sliding the spare in before the line bent out of shape.
Bacteria have a repair job like that in their DNA. The DNA is the rail line, a tiny guide marks the address, and a spare piece with matching ends is the new rail. People had thought this kind of swap needed a full crew of helpers to turn up together. If one worker could start it alone, far more yards might be fixable.
In a soil bacterium called S. virginiae, the kit already looked stripped back. Its main worker, SviCas3, was small for this sort of job. Using short DNA rings that carried both the painted address and the spare piece, the cell removed or replaced several chosen stretches, then managed another round after the first edit had already gone in.
Then the tool was moved into E. coli, and the surprise got bigger. More of the usual crew could be left behind, yet the swap still happened. With the address clearly marked and the matching spare in place, SviCas3 on its own still helped the cell cut out a stretch or slot a new one in. It looked like one compact rail worker was enough to wake the yard's own repair team.
The same lean setup also worked in Corynebacterium, which can be awkward to edit because cells may not survive a better known cutter. The swaps happened there too, though not as neatly, and some colonies showed a mix of old and new track. It also coped with different end patterns and different spare lengths, while quick checks at tested nearby spots did not show stray changes.
Some of the puzzle is still unsolved, and the safety look was only a partial one. But the picture has changed all the same. What seemed like a job for a whole rail crew can, in some bacteria, begin with one compact worker, one painted address, and one matching spare piece. That is the bit people did not expect.
